Another thing to consider, especially if you have a lot of woodworking experience, is while your subflooring is off, take the time to ensure all of your joists are perfectly in line with each other. A laser level helps, but you can do it with a regular one. There's often a little crown or dip on at least one joist, and having a perfectly flat set of joists makes for a much easier floor to tile on.
As CX was indicating...ALL layers of ply should go across the joists...you do NOT orient them at 90-degrees to each other.
If you want to keep your total floor assembly height down, you can save about 1/2 the height of that 1/4" cbu by using Ditra.
